Synthesis of a Diapocynin Prodrug for Its Prolonged Release from Zwitterionic Biodegradable Nanoparticles

Today, neurodegenerative diseases are affecting more and individuals. A typical therapeutic target to treat the progression of symptoms related these is proinflammatory microglia. Diapocynin, dimeric version apocynin, often drug choice. However, suitable carriers for its controlled delivery currently lacking in clinics. In fact, high hydrophilicity hampers development a formulation enabling sustained release biological environment. this work, possibility modifying diapocynin explored order synthesize prodrug that can be chemically incorporated into biodegradable zwitterionic polymer nanoparticles (NPs). These NPs synthesized via combination ring opening polymerization reversible addition–fragmentation chain transfer (RAFT) polymerization, allowing incorporation desired number units carrier thus set dosage priori. The chemical binding avoids premature compared physical loading inside similar nanovectors. With strategy, achieved than 9 days, paving way therapy with low repeated administrations.
